Begomoviruses are significant constraints in horticultural crops in India. As several begomovirus are known to infect a single crop species with similar symptoms, it is necessary to have a sensitive and specific detection for each begomovirus species. However, rapid and specific diagnostic methodology for differentiating these begomovirus species is not available. Technology Details:
We have developed a differential PCR assay based on 3’ polymorphic primers for differentiation of begomovirus species. The assay was validated detecting six begomovirus species infecting Tomato and Chilli in India. A patent has been filed for this invention. The assay methodology has been utilized to develop six easy to use PCR kits for each of the following begomoviruses: tomato leaf curl New Delhi virus, tomato leaf curl Bangalore virus, tomato leaf curl Palampur virus, tomato leaf curl Gujarat virus, tomato leaf curl Joydevpur virus and chilli leaf curl virus. The easyPCR kit contains has been simplified in a single tube containing Taq DNA polymerase, buffer, primers and dNTP. The kit has been designed with the ease of use as only the test DNA to be added in the supplied tube to perform the PCR. These easy PCR kits will be useful in screening begomovirus resistance in the plant genotypes as well as in identifying the distribution pattern of these begomoviruses in the different cropping regions.
